Orexin and psychiatric disorders

چکیده
Orexin-A and Orexin-B (also known as hypocretin 1 and 2) are, respectively, 33and 28-amino acid residue peptides that activate a G-protein-coupled ‘orphan’ receptor, i.e. which has no known ligand.1 Immuno-cytochemical studies show that orexin-positive neurons are located in the lateral hypothalamic area and arcuate and perifornical nuclei.2 They are also found in various sites beyond the hypothalamus such as the thalamic paraventricular and reuniens nuclei, locus coeruleus, nucleus of the solitary tract, raphe and septal nuclei.

Orexin and Circadian Influences in Sleep and Psychiatric Disorders: A Review of Experimental and Computational Modelling Studies
Psychiatric disorders such as unipolar depression have complex pathologies, which include disruptions in circadian and sleep-wake cycles. At the neurochemical level, psychiatric diseases can also be accompanied by changes in neuromodulator systems such as orexin/hypocretin and the monoamines. Indeed, for decades the monoamine hypothesis of depression has been instrumental in driving discoveries...
متن کاملOrexin antagonists for neuropsychiatric disease: progress and potential pitfalls
The tight regulation of sleep/wake states is critical for mental and physiological wellbeing. For example, dysregulation of sleep/wake systems predisposes individuals to metabolic disorders such as obesity and psychiatric problems, including depression. Contributing to this understanding, the last decade has seen significant advances in our appreciation of the complex interactions between brain...
متن کاملTargeting the orexinergic system: Mainly but not only for sleep-wakefulness therapies
Orexin receptors belong to the big family of G protein coupled receptors (GPCRs) that constitute the main targets in the modern pharmacological approaches. Although the orexinergic system is involved in a variety of processes, treating sleep-wakefulness disorders such as narcolepsy and insomnia, remains the main therapeutic implication of targeting orexinergic receptors. After novel advances, s...
متن کاملOrexin signaling mediates the antidepressant-like effect of calorie restriction.
During periods of reduced food availability, animals must respond with behavioral adaptations that promote survival. Despite the fact that many psychiatric syndromes include disordered eating patterns as a component of the illness, little is known about the neurobiology underlying behavioral changes induced by short-term calorie restriction. Presently, we demonstrate that 10 d of calorie restri...
متن کاملبررسی نقش ارکسین A در دوره بارداری و شیردهی و سندرم مرگ ناگهانی نوزاد: مروری سیستماتیک
Background and Objectives: Adipose tissue plays a role in releasing hormones including orexin, that regulates energy metabolism, nutrition, body weight, control system, autonomic, sleep, insulin sensitivity, breathing, drug addiction ,and reproductive axis. The pregnancy is associated with a series of metabolic changes that provide energy and nutrients for mother and fetus. Orexin increases ins...
